We constructed the Greenland Soil Microbiome Gene Catalog (GSMGG), comprising over 468 million non-redundant genes predicted from 5,307 high- to medium-quality metagenome-assembled genomes (MAGs) recovered from 283 active-layer soil samples across Greenland.

This catalog represents one of the most comprehensive gene-level resources available for Arctic terrestrial microbiomes. All genes were systematically annotated across multiple functional dimensions, providing deep insights into microbial adaptation to extreme cold and acidic conditions, biogeochemical cycling, greenhouse gas dynamics, and untapped biotechnological potential.

Functional Annotation Highlights

General Functional Annotation

Comprehensive assignment using KEGG Orthology (KO), COG, and eggNOG databases, enabling pathway reconstruction, metabolic network analysis, and comparative functional profiling across phyla and environmental gradients.

Carbon, Nitrogen, Sulfur, and Methane Cycling Genes

Detailed cataloging of genes involved in:

  • Carbon fixation, degradation, and recalcitrant compound breakdown
  • Methane production (methanogenesis) and oxidation
  • Nitrogen fixation, nitrification, denitrification, and dissimilatory nitrate reduction
  • Sulfur oxidation and sulfate reduction

These annotations highlight the microbial drivers of greenhouse gas (CO₂, CH₄, N₂O) fluxes and their responses to rapid Arctic warming.

Carbohydrate-Active Enzymes (CAZymes)

High-resolution annotation of cellulases, xylanases, laccases, peroxidases, glycosyltransferases, and other CAZy families.

Patterns show clear increases in lignocellulose-degrading capacity along vegetation succession and soil development gradients — revealing a rich reservoir of cold-adapted polysaccharide-degrading enzymes with potential biotechnological applications.

Cold Adaptation and Extreme Environment Genes

Enrichment of genes supporting survival in sub-zero temperatures and freeze-thaw cycles, including:

  • Lipopolysaccharide (LPS) biosynthesis and membrane fluidity maintenance
  • DNA mismatch repair and oxidative stress resistance
  • Cold-protective secondary metabolites (hopene, flexirubin, glycopeptidolipids)
  • Viral auxiliary metabolic genes (AMGs) that enhance host freeze tolerance

Biosynthetic Gene Clusters (BGCs)

Detection of 14,592 BGCs across major classes (NRPS, terpenes, etc.).

Strikingly, >99% show no close homology to known BGCs in MIBiG or global BGC atlases — positioning Greenland soils as one of the richest untapped sources of novel bioactive compounds on Earth.

Extremely High Novelty

Antibiotic Resistance Genes (ARGs)

Identification of 1,705 ARGs conferring resistance to 28 antibiotic classes, dominated by multi-drug efflux systems (ABC, MFS, RND families).

Provides critical baseline data on the environmental resistome in a remote, minimally anthropogenically impacted Arctic region.

Additional Functional Layers

  • CRISPR-Cas systems (including many unclassified types) and anti-CRISPR proteins
  • Iron metabolism, hydrogenase, and trace gas utilization genes
  • Viral auxiliary metabolic genes influencing host nucleic acid, carbohydrate, vitamin, and amino acid metabolism
